Exosome and Ago2 proteins were isolated from organic-phenol fraction during total RNA isolation using miRNeasy kit and resolved in 8M urea. Protein concentrations were quantified using Bradford assay and total of twenty microgram protein from each group were separated in 12 % SDS-PAGE, transferred nitrocellulose membrane and incubated with specific antibody (CD63 and Ago2). Followed by HRP-conjugated secondary antibody and detected using chemiluminescent substrate. Western-blot results indicate the specificity of exosome and Ago2 protein isolation from follicular fluid (FF) and blood plasma (BP) as indicated in the corresponding figures
