In situ hybridization was performed with cenT and cenC padlock probes labeled with dinitrophenyl-11-dUTP and digoxigenin-11-dUTP, respectively. Probes were visualized with fluorescein-labeled anti-dinitrophenyl Fab fragment (Molecular Probes) and rhodamine-labeled anti-digoxigenin Fab fragment (Boehringer-Mannheim GmbH). The green and red signals were quantitated from 10 metaphase images, where the background was subtracted and the strongest signal was set to 100%.
