Figure Legend Snippet: (A) SMAUG1 phosphorylation status in HEK293 cells was determined using a Mn +2 Phos-tag SDS-PAGE gel. The phosphorylation status of SMAUG1 was determined through comparison of gel migration of lambda phosphatase-treated protein samples compared to untreated protein samples. Treated and untreated samples from whole-cell lysates were subjected to SDS-PAGE on 8% polyacrylamide gel containing 0 μM (no Mn2 + –Phos-tag) or 25 μM Mn2 + –Phos-tag, followed by immunoblotting with anti-SMAUG1, anti-AKT and anti-phospho-AKT Ser 473 . (B) Anti-myc co-immunoprecipitations with FLAG-tagged wild-type SMAUG1 and myc-tagged 14-3-3γ from co-transfected HEK293 cells. SMAUG1 is present in the bound fraction only when 14-3-3γ is co-expressed in the cells; β-actin was used as a negative control. (C) Anti-myc co-immunoprecipitations with FLAG-tagged SMAUG1 that has Ser to Ala mutations at positions 168, 254, 580 and 665 within four 14-3-3 binding motifs. Mutant FLAG-tagged versions of SMAUG1 were co-transfected with myc-tagged 14-3-3γ in HEK293 cells. Detection of SMAUG1 in the bound fraction decreases as the number of mutated 14-3-3 binding motifs increases. This indicates that 14-3-3γ interacts with SMAUG1 in a multivalent manner.
