Olorofim belongs to a new class of antifungal agents called the orotomides, which inhibits fungal pyrimidine biosynthesis. Olorofim inhibits dihydroorotate dehydrogenase, an enzyme of de novo pyrimidine biosynthesis, leading to the rapid depletion of uridine 5′-monophosphate (UMP), uridine diphosphate (UDP), and uridine triphosphate (UTP). The reduction in UTP levels can cause a decrease in cell wall biogenesis, protein glycosylation, and synthesis of DNA, protein, and phospholipid. In Sporothrix yeast cells, the inhibition of pyrimidine synthesis by olorofim may induce cell cycle arrest and affect the fungal cell wall, resulting in cell lysis.
