Combination of OMT and DG mitigates the DNFB-induced skin inflammation in mice. ICR mice were smeared with DNFB for five times and after first smearing, the mice were randomized and treated intraperitoneally with saline (model), or the indicated doses of OMT and/or DG daily for 14 days. Their body weights, skin inflammation and ear thicknesses were measured longitudinally. (a) The structures of OMT or DG. (b) The body weights of the different groups of mice. (c) Representative images of skin inflammation in different groups of mice (on day 14). (d) The inflammation scores of individual groups of mice. (e) The ear thicknesses. Data are representative images or expressed as the mean values of individual groups (n = 5 per group). The skin inflammatory scores and ear thicknesses of all ACD mice were significantly greater than the control while the skin inflammatory scores and ear thicknesses in all drug-treated groups were significantly less than that in the model group (statistical markers not shown in Figure 1(d) and (e)). Letters on the graphs indicate the following statistical significant group comparisons: aP< 0.05 vs. the control group; bP< 0.05 vs. the model group; cP< 0.05 vs. the DEX 2 mg·kg−1 group; dP< 0.05 vs. the combination (OMT+DG, 45 + 45 mg·kg−1). Figure 6. OMT exhibited anti-cancer function in CC cells by upregulating miR-197-3p to downregulate RRM2. (a-b) Transfection efficiencies of anti-miR-197-3p (a) and si-RRM2 (b) were examined by RT-qPCR and western blot, respectively. (c-l) 6 mg/mL-treated CaSki and SiHa cells were transfected with anti-miR-197-3p, anti-miR-197-3p+si-RRM2, or relative controls. (c) CCK-8 assay was performed for viability detection. (d-e) The examination of cell proliferation was conducted using colony formation assay (d) and Edu assay (e). (f-h) Cell apoptosis analysis was implemented through flow cytometry (f) and western blot (g-h). (i-l) Wound healing assay (i-j) and transwell assay (k-l) were performed to assess cell motility. If we only consider the concentration of alkaloidal matrines as bioactive materials, the contents of matrine and oxymatrine in some products among the tested biopesticides were assumed to be suffi- cient as crop protection agents. However, if we consider the practical use guideline of biopesticide, the concen- tration of the active ingredient would be expected to be lower than EC50 and LD50 level, because the general guideline for field application was suggested be 100 - 1000 folds diluted with water. In a survey of matrines present in biopesticides from S. flavescens, the concentration of bioactive materials showed wide ranges and these results would be related to the crop protection efficiency. Thus, the quality control of the commercial bio- pesticide should be performed.
