Fig. 1. <t>H6PD</t> and regulation of glucocorticoid metabolism with enz resistance. (A) Effects of enz treatment on H6PD, GR, and 11-HSD2 in the LAPC4 CRPC model as assessed by immunoblot. (B) NADPH generation in LAPC4 cells with enz treatment. NADPH was assessed using a bioluminescence assay in ER extracted from control and enz-treated cells. (C) H6PD and 11-HSD2 protein in tissues from enz-treated patients with prostate cancer. Bar graphs represent band intensity; three paired tissues from three patients with prostate cancer were analyzed. (D) Effects of enz on H6PD and 11-HSD2 protein with ex vivo treatment of 12 clinical tissues from patients with prostate cancer. For all panels, unless otherwise noted, error bars represent the SEM, and P values were calculated using unpaired two-tailed t tests.

Fig. 1. Effect of starvation on G6PT, <t>H6PD</t> and 11bHSD1 enzyme activities in rat liver microsomes. (A) Activity of the G6PT–H6PD–11bHSD triad. Microsomes were incubated at 37 C in KCl/MOPS buffer in the presence of 10 lM cortisone and 50 lM G6P at a protein concentration of 0.5 mg/ml for 5, 10 and 20 min. The reaction was terminated by addition of ice-cold methanol. The ratio of cortisone– cortisol conversion was analyzed by HPLC (see Section 2). (B) Enzyme activity of H6PD. Microsomes were incubated at room temperature in KCl/MOPS buffer in a fluorimetric quartz cuvette at a protein concentration of 1 mg/ml. Dehydrogenation of G6P was monitored via NADPH formation (extinction and emission wavelengths at 350 and 460 nm, respectively) after the subsequent addition of 2 mM NADP+, 100 lM G6P and 1% Triton X100. Pulse addition of NADPH was done for calibration. All experiments were elaborated in triplicates. The quantified results are shown as mean values ± S.D. Statistical analysis was performed with the Tukey–Kramer Multiple Comparisons Test.

Fig. 1. Effect of starvation on G6PT, H6PD and 11bHSD1 enzyme activities in rat liver microsomes.

A schematic illustration of the working model of this study . The present study demonstrated that hypoxia acclimation-associated inflammatory cytokines upregulate the hexosamine biosynthetic pathway (HBP) and activate G6PDH by increasing the O-GlcNAcylation, which improves pentose phosphate pathway (PPP)-mediated redox homeostasis, and alleviates acute cardiac I/R injury.
