Figure Legend Snippet: (A) A DNA fragment was methylated in vitro using the HpaII DNA methyltransferase (HpaII MT) and subsequently integrated into C. neoformans. gDNA extracted from isolates that correctly integrated the fragment, was digested with the restriction enzyme HpaII. To test the maintenance of the in vitro-methylated 5mC, qPCR was performed on region spanning the restriction sites. To test the ability 5mC to spread, gDNA was digested with either Hpy99 or BstUI and qPCR was performed across these sites. White and black lollipops represent unmethylated and methylated sites, respectively.
